What helped the game spread from one data center to another was the fact that the source code was freely available. It was created in 1977 by Willie Crowther and Don Woods and quickly became a phenomenon in those pre-GUI, pre-Internet years. They’re the starting point for Colossal Cave Adventure, the first major computer game. Sound familiar? If you’re an old-school gamer, you’ve probably seen these words hundreds of times. Many elements of the original game have survived into the present, such as the command ' xyzzy', which is now included as an Easter Egg in games such as Minesweeper" (Wikipedia article on Interactive fiction, accessed 04-15-2009).You are standing at the end of a road before a small brick building.Īround you is a forest. The popularity of Adventure led to the wide success of interactive fiction during the late 1970s and the 1980s, when home computers had little, if any, graphics capability. The game has since been ported to many other operating systems, and was included with the floppy-disk distribution of Microsoft's MS-DOS 5.0 OS. "In early 1977, Adventure spread across ARPAnet, and has survived on the Internet to this day. ( See the original source code) The program required about 60K words (nearly 300KB) of core memory in order to run, which was a significant amount for PDP-10/KA systems running with only 128K words." (Wikipedia article on Colossal Cave Adventure, accessed 04-14-2009). ![]() ![]() "Crowther's original game consisted of about 700 lines of Fortran code, with about another 700 lines of data, written for BBN's PDP-10. The game was renamed Colossal Cave Adventure, as it was based on part of the Mammoth Cave system in Kentucky. When a hormone binds to the receptor, the G-protein is activated by binding guanosine triphosphate, or GTP, in place of GDP. When a hormone is not bound to the receptor, the G-protein is inactive and is bound to guanosine diphosphate, or GDP. When a hormone binds to its membrane receptor, a G-protein that is associated with the receptor is activated G-proteins are proteins separate from receptors that are found in the cell membrane. One very important second messenger is cyclic AMP (cAMP). An enzyme called phosphodiesterase breaks down cAMP, terminating the signal. cAMP is a second messenger that mediates a cell-specific response. Hormone binding to receptor activates a G-protein, which in turn activates adenylyl cyclase, converting ATP to cAMP. The amino acid-derived hormones epinephrine and norepinephrine bind to beta-adrenergic receptors on the plasma membrane of cells. The hormone that initiates the signaling pathway is called a first messenger, which activates a second messenger in the cytoplasm, as illustrated in Figure 18.6. In this way, nothing passes through the cell membrane the hormone that binds at the surface remains at the surface of the cell while the intracellular product remains inside the cell. Binding of these hormones to a cell surface receptor results in activation of a signaling pathway this triggers intracellular activity and carries out the specific effects associated with the hormone. Unlike steroid hormones, lipid insoluble hormones do not directly affect the target cell because they cannot enter the cell and act directly on DNA. Lipid insoluble hormones bind to receptors on the outer surface of the plasma membrane, via plasma membrane hormone receptors. In this way, the steroid hormone regulates specific cell processes as illustrated in Figure 18.5.Īmino acid derived hormones and polypeptide hormones are not lipid-derived (lipid-soluble) and therefore cannot diffuse through the plasma membrane of cells. This protein can be used either to change the structure of the cell or to produce enzymes that catalyze chemical reactions. This, in turn, determines the amount of corresponding protein that is synthesized by altering gene expression. The hormones and receptor complex act as transcription regulators by increasing or decreasing the synthesis of mRNA molecules of specific genes. The cell signaling pathways induced by the steroid hormones regulate specific genes on the cell’s DNA. The steroid hormones pass through the plasma membrane of a target cell and adhere to intracellular receptors residing in the cytoplasm or in the nucleus. At the target cell, the hormones are released from the carrier protein and diffuse across the lipid bilayer of the plasma membrane of cells. Once outside the cell, they bind to transport proteins that keep them soluble in the bloodstream. Lipid-derived (soluble) hormones such as steroid hormones diffuse across the membranes of the endocrine cell. Depending on the location of the protein receptor on the target cell and the chemical structure of the hormone, hormones can mediate changes directly by binding to intracellular hormone receptors and modulating gene transcription, or indirectly by binding to cell surface receptors and stimulating signaling pathways. Receptor binding alters cellular activity and results in an increase or decrease in normal body processes. When the number of receptors decreases in response to rising hormone levels, called down-regulation, cellular activity is reduced. In up-regulation, the number of receptors increases in response to rising hormone levels, making the cell more sensitive to the hormone and allowing for more cellular activity. Additionally, the number of receptors that respond to a hormone can change over time, resulting in increased or decreased cell sensitivity. The number of receptors that respond to a hormone determines the cell’s sensitivity to that hormone, and the resulting cellular response. Cells can have many receptors for the same hormone but often also possess receptors for different types of hormones. For example, thyroid hormones act on many different tissue types, stimulating metabolic activity throughout the body. Receptors for a specific hormone may be found on many different cells or may be limited to a small number of specialized cells. In this way, even though hormones circulate throughout the body and come into contact with many different cell types, they only affect cells that possess the necessary receptors. Hormones mediate changes in target cells by binding to specific hormone receptors. Discuss the role of different types of hormone receptors. ![]() ![]() ![]() In addition to proof of identity, such as passport, birth certificate or adoption certificate, and proof of your address, or a letter from your university or college confirming your place, and bring it along with your other identity and address verification documents to your Over 16's don't need to be accompanied by a parent but need to bring in proof of their identity and proof of their address. Arguably, there may be even more then you imagine, since the tier of a deck is changed by the popularity of other decks. There usually is a few more S tier (Or T1) decks lying in wait in any given patch, totally undiscovered. S tier Control v Control metas fucking suck, games take forever, and they're incredibly unsatisfying. I never ever want to go back to the Control Warrior meta where me and my opponent sat there for 30+ turns tanking up at eachother to see who dies to fatigue first through our 250 armor, for short 1.5h games of glaring at eachother, unable to play our threats because of Shieldslam and execute. These "shitty tools" people complain about are already pretty effective in an Aggro Meta, and very few people who are pushing for control tools to be better have, imo, actually experienced a control-grinder meta. Honestly, I'm not even sure why the opinion that "Well Control decks should beat." always pops up. Miracle Rogues are usually a hybrid between Combo and Tempo, both of which are generally going to push control away. (People often don't recognize Tempo decks as their own thing and confuse them with Aggro, decks like Face Hunter, Handlock, Cubelock, Big Easy Priest, etc, are all Tempo Decks, and control struggles with these decks because their gameplan is basically "an inevitable victory" on a clock.) Control decks react to your opponents plays, you cannot react to an opponent who plays solitaire for 7 turns before hitting your face for 1000 out of hand.Ĭontrol decks also often struggle to deal with Tempo decks since they're designed to assert dominance over the game-state rather the the Board. As anyone reading this already know, that doesn't happen very often these days.Ĭontrol decks get dunked on by Combo. Then you could save your big healing turn for when your opponent has to spend a turn making clearing your board or drawing more gas. This would be balanced if opponents could run low on resources. The best I can think of is Holy Maki Roll, but that again ends up costing you your entire turn for a big heal. And there are no options outside of Reno Jackson to heal a large amount relative to mana spent (Vampiric Blood in DK is often identical to "gain 10 armor" because of this). It's absurd that other classes get consistent options to deal huge damage to face while drawing and building board, but powerful healing is usually conditional and costs so much you rarely have the mana to impact the board state. Meanwhile control gets premium defensive options like "4 mana lifesteal Fireball that targets only minions" or "put 2 damage life steal dot on enemy board" and hope most of them have 2 hp and/or your opponent doesn't have an aoe attack boost or burn and/or the board isn't mage skeletons and you didn't even manage to heal off your opponent's board. ![]() Resources Fireside Gatherings Related Subreddits Chat (Discord) Sticky Threads & Guides Community Sites Tools Icons made by Daniel Bruce, Freepik, Icomoon, Icon Works, and Bogdan Rosu from are licensed by CC BY 3.0. ![]() The plot involved luring Caesar to Pompey’s theatre, where gladiatorial games were held in his honor. They sought to liberate Rome from Caesar's rule. ![]() The members of the plot called themselves the liberators. The guiding spirit behind the conspiracy was Cassius, a leading Roman senator. However, many of the facts of the assassination have been established with a great deal of certainty. None of the sources are contemporary, and they often contradict each other. The sources on the assassination and the conspiracy could be best described as imperfect. The members of the conspiracy were all prominent Romans who knew Caesar. It seemed that his position was beyond challenge, but this situation changed on the Ides of March 44 BCE.īy March 44 BCE, members of the Roman elite conspired to remove Caesar from power. This meant that, along with the army under his control, he was granted extraordinary powers. He had been elected by the Senate as dictator initially for five years, but later for life. By this time, Caesar was the most powerful man in Rome. The conflict continued to rage over the Mediterranean for several years.Ĭaesar emerged victoriously and by 44 BC after his decisive victory at the Battle of Mutina. Mark Anthony ruled in Caesar’s name in Rome and Italy. Caesar defeated his enemies at Pharsalus (Greece), but this did not end the war. Pompey the Great defended Rome alongside the Senate. In response, Caesar advanced on Rome to secure his political position and started a civil war between himself and the Roman senate. The Senate and Pompey saw an opportunity to oust Caesar from power and threatened him with criminal prosecution. He then used his legions to raid the people in Germany and Britain.Īfter the death of Crassus, the First Triumvirate broke down. Caesar then embarked on a ten-year campaign to gradually conquered this region. At the time, Gaul covered most of France and Belgium. Caesar lead these legions into Gaul to pacify this region. Under this arrangement, Caesar was able to secure election as consul and to become the commander of several Roman legions. This agreement allowed the three men to secure their political goals and become, in essence, the de-facto government of the Republic. Caesar entered into an informal political arrangement with Crassus and Pompey, known as the First Triumvirate. He was a charismatic leader and became one of the best-known figures on the Roman political scene. Upon the death of Sulla, he returned to Rome and became one of the leaders of the popular party in Rome. Caesar quickly rose through the ranks of the military and distinguished himself. Caesar sided with Marius, but when the war shifted in favor of Sulla, Caesar was forced to flee Rome and join the army to avoid execution. Additionally, the death of his father left him somewhat unprotected.Ī bloody civil war between Marius and Sulla forced Caesar to pick sides. We know little about Caesar's childhood, but Rome was unstable during his teenage years. However, by the time of Caesar’s birth, their fortunes had declined, and they were no longer particularly prominent. ![]() The assassination of Julius Caesar ultimately started a civil war that ended with the destruction of the Roman republic.Ĭaesar was born to a patrician Roman family who had once been very influential in the Republic. Finally, some were motivated by basic human emotions - personal vengeance. Second, they tried to prevent him from becoming king and destroying the Roman Republic. Why was Caesar murdered at the hands of his fellow citizens? Caesar was killed for three reasons: First, the conspirators wanted to halt his power growth. On the Ides of March, Caesar was stabbed to death by these men who wanted to preserve the Roman Republic. However, Caesar's actions to marginalize the Republic and seize sole control of Rome angered many Romans, including men he not only knew but considered friends. ![]() But he also was quickly dissolving the Roman Republic and creating what would become the Imperial system. During his reign, Caesar greatly expanded the Roman Republic, and his conquests changed Europe's future. He had taken control of Rome not just through his military might but with charisma and political savvy. Caesar had become one of the best-known and prominent figures of the ancient world at this point in his life.
